It is possible that Silas is the Cure, and Shane wants his family to be brought back with a spell, only an Immortal Sorcerer like Silas can do once he regains his magical powers.
When Damon tries to unsire Charlene , the witch made him kill twelve innocent people to broke the sire bond. Only that didn't work, because there's no magic to broke a sire bond between two vampires, so when he learn that, he learn at the same time that the spell the witch made him do, by killing 12 innocent people, was actually to bring to her a sort of black magic. So I think that when Klaus kill all the twelve hybrid, exept tyler, that was to made Shane (?Silas?) a black witch.
Then why Shane wants the cure, I think its to kill Klaus. Because with Klaus the more powerfull creature in Mystic Falls Shane (?Silas?) can't be completly powerfull.
The "Cure" or whatever is really is, I don't think is what Shane truly after. He did say he's not interested in the cure. From everything he said, and him teaching Bonnie to do "Expression," which is apparently very dark magic, I'm guessing he's trying to resurrect the dead.
Or it's possible the what the cure does is actually killing vampires. The first time the cure is mentioned, Rebekah's hunter lover, Alexender said it's an ultimate weapon that no vampire can survive. Vampires are undead creature, meaning they're alive and dead at the same time, maybe the cure will take away one part, making vampires really dead.
Or it is evidently possible that Shane is fully working to free Silas, that everything he has said is a lie and that he is a good actor, that Silas could actually be the cure to Vampires being the original Immortal.
I don't know. In 4x15 Ester said that as the witch who turned her children into vampires, she had the power to turn them into humans. The way she said it led me to believe that she was the only one who could turn them back. Kind of like the Bennett witches were the only ones that could open her casket.
Could we possibly see Ester return once more? From the looks of it in 4x15 she was going to use a spell to turn them human which goes along with what Shane was saying about the hunters mark and the spell he needed.

In the last episode we saw April open Rebekah's coffin and watch klaus kill the hybrid, she was wearing the braclet Jeremy gave her so when she found Caroline so she couldn't be compelled, and the fact that it was her father that killed the council and himself and shane knew her father, does anyone else think that see might seek revenge for her father, or either klaus/shane begins using her as a tool, shes been a quiet person for saying her father killed the council, i think she will turn to shane after discovering about vampires and as a human she will either be sacrificed or be a vessel. I think april is going to be a part of the storyline.            Any other thoughts on april??
Silas is a warlock who lived who-knows-how-much times before. He wanted to do a spell that grants him immortality. Qetsiya, i witch who loved him, did the spell together with him and he became immortal. Silas then fell in love with another woman and wanted to do the spell on her so that she would be immortal to but Qetsiya felt betrayed and became angry so she killed the other woman and buried Silas alive. http://vampirediaries.wikia.com/wiki/Silas
i don't know if Qetsiya also became immortal but it is said that Silas is still in his grave waiting for someone to find him and excavate him so he can take revenge on the world. Maybe Professor Shane wants the cure for bringing back Silas, don't know why.

I think you mean 3x15. I think what she meant was that it was her responsibility on one hand and that she was the only one with the knowledge and power to do it. If you'll remember from season 1 they needed Emily's Grimoire to open the tomb because they needed the right spell since every spell is unique unto itself. I think this is what Esther possessess, the knowledge of how the spell was done and thus how to undo it and since her grimoire has most likely already turned to dust (or it wasn't her spell to begin with). However we may indeed see the return of Esther, her storyline was left somewhat unfinished, although I'm wondering if they haven't just decided to drop her since she wasn't the most popular villain ever with the fanbase, either that or they're probably gonna bring her back at the most shocking moment possible.
