The relationship between John Gilbert and Isobel Flemming. They used to be in love before Isobel left Mystic Falls. They share a daughter together, Elena Gilbert.

Early life

When they were in High School, John and Isobel dated and slept together. This resulted in a child, Elena, who was delivered by John's brother, Grayson Gilbert. No one knew who the real father was.

At some point, John told Isobel about vampires, something she started obsessing over. He also gave her his Gilbert ring, wanting to protect her.

Finally, in 2007, she expressed a desire to meet a vampire, and he sent her to Damon. He did not know of her true motives, and later confessed to Katherine (believing her to be Elena) that had he known what she wanted to do, he would never have sent her to him. He also admitted that part of the reason why he hates vampires, is because of what being one did to Isobel.

Season 1

In season 1, it is revealed that Isobel and John met when they were teens. While John fell in love with Isobel, she never felt the same in return. However, she was interested in John's knowledge about supernatural creatures, and therefore she stayed with him. A flirt was introduced between the two of them and Isobel got pregnant with John's child. She gave birth to their daughter and thanks to John, Isobel was able to deliver at the hands of Grayson Gilbert, John's brother and doctor (who didn't know John was the father). According to John himself, he was with Isobel during the delivery, and he later told Elena how heart-broken she had been while giving birth. Isobel decided to give up the baby because she and John were simply too young to take care of it themselves. Grayson and his wife, Miranda, took care of the little girl instead and became her adoptive parents. 

John and Isobel's relationship didn't proceed further after that, they went different ways and chose different lifestyles. While Isobel started studying supernatural creatures, John had a very disdainful attitude towards vampires. Especially after Isobel herself became a vampire in 2007, her personality changed a lot which made John's contempt even worse, because he saw what vampirism did to her. The only thing they shared was the want of protecting Elena from the supernatural creatures, especially Stefan and Damon Salvatore. 

While Isobel was married to Alaric Saltzman, she never told him about her past with Elena (and John).

Season 2

When Isobel died in "Know Thy Enemy", it was never revealed how John handled it, although it seems he did not really care. Possibly the Isobel he got to know in his teens was so different from the vampire Isobel that he thought she was already long gone.
